Therefore, the objective of this study was to evaluate N, N'- diformyl urea (2%) as a strategy to mitigate water stress in passion fruit seedlings. The variables analyzed included the number of true (unfolded) leaves, stem diameter, dry weight of roots and shoots, as well as proline determination. The study was conducted using a split-plot design with a randomized complete block arrangement. The main plots corresponded to the soil substrate moisture levels in each nursery bag: optimal conditions at field capacity (FC), initial permanent wilting point (PWP), and an intermediate condition between the two. The main plots corresponded to the soil substrate moisture levels in each nursery bag: optimal conditions at field capacity (FC), initial permanent wilting point (PWP), and an intermediate condition between the two. Plants subjected to severe water stress (PMP) and treated with diformyl urea at a concentration of 1.5 cc L-¹ exhibited similar behavior to the control treatment under field capacity (CC) conditions, with 7.83 leaves and a stem diameter of 4.25 mm, compared to 7.58 leaves and a stem diameter of 4 mm in the control. Thus, applying diformyl urea could help plants better endure water deficit conditions, improving their capacity to adapt to drought scenarios. The proline concentration in yellow passion fruit plants under severe water stress (PMP) and treated with diformyl urea at 1.5 cc L-¹ was higher than in other treatments, reaching 10.61 µM. These findings suggest that the upregulation of proline biosynthesis in plants exposed to this compound may confer enhanced physiological resilience to water stress tolerance in yellow passion fruit.
Effect of heavy metals on strawberry (Fragaria × ananassa). A review http://www.scielo.org.co/scielo.php?script=sci_arttext&pid=S2011-21732025000300003&lng=pt&nrm=iso&tlng=pt ABSTRACT Despite their high nutritional properties, strawberries are classified as the most contaminated fruits. Their contaminants include heavy metals (HM) such as cadmium (Cd), lead (Pb), mercury (Hg), chromium (Cr) and the metalloid arsenic (As). The sources of these contaminants are water, air and soils of geogenic and anthropogenic origin, the last of which is the biggest contributor. This situation poses a health risk to humans. Between 3 and 16% of agricultural soils are affected by excessive levels of HM, especially those near industrial and mining areas, as well as those with high concentrations of pesticides and fertilizers. There is a strong correlation between the HM content in the soil and the HM content in the strawberry plants, with the greatest accumulation of HM in the root system. HM are absorbed by the roots but also aerially by the leaves, and they are also retained in the cuticle wax of the fruits. Excess HMs, especially Cd, in strawberry plants manifest themselves in a decrease in biomass, chlorophyll, stomatal conductance, growth hormones, and leaf area, as well as in oxidative damage, lower photosynthetic efficiency, and changes in the accumulation and redistribution of mineral elements. Strategies for reducing contamination in strawberry plants include the application of silicon, humic acid, melatonin, abscisic acid, or Bacillus spp., as well as the use of nanoparticles (e.g. SiNPs), inoculation with arbuscular mycorrhizal fungi, biochar amendments, and planting HM hyperaccumulators, among others.
Proteomics reveals an association between water stress severity in potato and photosynthetic proteins http://www.scielo.org.co/scielo.php?script=sci_arttext&pid=S2011-21732025000300004&lng=pt&nrm=iso&tlng=pt ABSTRACT Water stress is among the most critical factors limiting plant performance. Proteomics serves as a powerful tool for assessing changes in protein expression and identifying mechanisms underlying drought stress tolerance in plants. This study aims to identify proteins associated with water stress severity in potato (Solanum tuberosum L.) and to characterize their differential expression as potential defense mechanisms against stress. Potato plants at the tuber differentiation stage were subjected to mild, moderate, and severe water stress by withholding irrigation. Gas exchange measurements and proteomic analyses were conducted at each stress level. For protein extraction and proteome profiling using two-dimensional polyacrylamide gel electrophoresis (2D-PAGE), leaf samples were collected from plants under different stress conditions and from well-watered controls. A total of 74 protein spots were detected, and proteins associated with water stress were selected based on correlation analysis between their relative abundance and leaf water potential. Seven proteins were identified via mass spectrometry. Increasing water stress severity was associated with a decline in gas exchange and an increase in water use efficiency. Notably, an increase in the relative abundance of Rubisco activase, Photosystem I subunit IV B reaction center, and the manganese-stabilizing protein of Photosystem II under water stress suggests a potential protective strategy for maintaining photosynthetic function. Conversely, three isoforms of cytochrome oxidase (COX) subunit I exhibited decreased expression, which may be linked to a heat dissipation mechanism aimed at mitigating reactive oxygen species (ROS) formation during water stress. The identification of these proteins provides a baseline to further understand water stress in potatoes.
Detection and molecular characterization of potato yellow vein virus (PYVV, Crinivirus flavisolani) in Solanum phureja from Chitaga, Colombia, and evidence of long-distance dispersal http://www.scielo.org.co/scielo.php?script=sci_arttext&pid=S2011-21732025000300005&lng=pt&nrm=iso&tlng=pt ABSTRACT The potato yellow vein virus (PYVV) is a Crinivirus transmitted by the whitefly Trialeurodes vaporariorum and is associated with significant economic losses in potato (Solanum spp.) cultivation in the Andean region. Until now, the presence of PYVV had not been reported in the department of Norte de Santander, Colombia. In this study, we report for the first time the detection of PYVV in Solanum phureja plants exhibiting typical vein yellowing symptoms in the locality of Chitaga. Leaf samples were collected and subjected to molecular analysis by RT-PCR, confirming the presence of the virus. The obtained amplicon was sequenced and deposited in GenBank under accession number PV593540. Phylogenetic analysis revealed that the sequence shares 98.8% nucleotide identity with other Colombian isolates of PYVV and clusters closely with sequences from the department of Nariño. This finding suggests a possible introduction of the virus into Norte de Santander through infected seed tubers. These results underscore the importance of implementing robust epidemiological surveillance systems and promoting the use of virus-free certified seed to prevent the dissemination of PYVV and other pathogens across potato-growing regions.
Evaluation of native isolates of Trichoderma spp. from Nariño (Colombia) as potential biological control agents against Rhizoctonia solani Kühn http://www.scielo.org.co/scielo.php?script=sci_arttext&pid=S2011-21732025000300006&lng=pt&nrm=iso&tlng=pt ABSTRACT Potato black scurf (Rhizoctonia solani Kühn) is one of the most important endemic diseases in potato crops in the Department of Nariño (Colombia). Its control includes integrated management, combining preventive and curative strategies, within which biological control with the use of natural antagonists such as the fungus Trichoderma sp., as a valuable resource. The objective of this work was to characterize native strains of Trichoderma that can potentially be used in the management of the potato black scurf. For this purpose, 68 soil samples were collected from potato and other crop producing areas. The morphological characterization of the isolates was performed on PDA medium, determining the daily mycelial growth rate, number of conidia and chlamydospores. Antagonistic activity against the target pathogen was assessed in vitro using dual-culture assays on both mycelial mats and sclerotia. Two commercially available strains were included as reference standards. Molecular identification was carried out by amplifying the internal transcribed spacer (ITS) region of genomic DNA, specifically targeting the ITS5 locus. Initially, eleven Trichoderma isolates were obtained. Among these, five strains -designated M9, M10, M16, M21, and M66- were molecularly identified as T. asperellum and preselected based on their consistent and effective antagonistic activity against R. solani. Afther 15 days of incubation on PDA medium, the isolates exhibited daily mycelial growth rates ranging from 14.3 to 19.7 mm d-1, concentrations of 3.24·10⁷ to 6.74·10⁷ conidia/mL, and of 7.0·10⁴ to 2.1·10⁵ chlamydospores/mL. The 11 isolates displayed varying degrees of inhibitory activity against R. solani. The five isolates mentioned above showed inhibition percentages of mycelial growth ranging from 41.8% to 64.46%, in comparison with two commercial strains, which exhibited inhibition rates of 20.7% to 38.7%. In assays conducted on R. solani sclerotia, the five T. asperellum mentioned and the commercial strains demonstrated inhibition of germination ranging from 85% to 100%. These five isolates were selected as promising candidates for further evaluation against R. solani, with the aim of developing a reliable and effective biological control agent for use by potato growers in the Nariño region.
Relationship between seedling traits and recovery time after transplanting in shallot grown from true seed http://www.scielo.org.co/scielo.php?script=sci_arttext&pid=S2011-21732025000300007&lng=pt&nrm=iso&tlng=pt ABSTRACT Shallot (Allium ascalonicum L.) seedlings frequently experience transplant shock, but the role of seedling traits in recovery dynamics remains unclear. This study investigated three seed-propagated varieties ('Lokananta', 'Merdeka F1', 'Sanren F1') under net-house conditions (June 2024, 5°S, 119°E) using ANOVA, regression, and correlation analyses. Results demonstrated that 'Merdeka F1' exhibited superior performance, with 28-42% greater biomass (2.36 g), enhanced root development (12.80 tips, 8.90 cm length), and faster recovery (5.77 days vs. 6.33-6.53 days) compared to other varieties. Pseudostem diameter emerged as the strongest predictor of recovery time (quadratic model: R 2=0.77; correlation: r=−0.86), with seedlings >4.5 mm diameter recovering ~1 day faster. Significant negative correlations were also observed between recovery day and biomass (r=−0.66) or leaf count (r=−0.27), while root traits showed minimal influence (R 2<0.04). These findings establish pseudostem diameter as a key selection criterion for transplant resilience and highlight 'Merdeka F1's genetic advantages. Future research should investigate optimal seedling age and biostimulant applications to enhance shallot transplant success.
Identification of new weed hosts of begomoviruses in crop-growing regions of Valle del Cauca, Colombia http://www.scielo.org.co/scielo.php?script=sci_arttext&pid=S2011-21732025000300008&lng=pt&nrm=iso&tlng=pt ABSTRACT Weeds and economically important crops (tomato, pepper and passion fruit) in Valle del Cauca are currently affected by viruses of the Begomovirus genus, including PLDV, RhGMCV, PRMV, PYMV, and CroGMV. This study aimed to identify weed species associated with crops of agronomic interest that may serve as natural reservoirs of these viruses. Leaf samples were collected from various weeds in crop fields across Candelaria, Cerrito, and Palmira, and total DNA was extracted. Virus detection was performed by PCR using both universal Begomovirus primers and specific primers for PLDV, RhGMCV, PRMV, PYMV, and CroGMV. The incidence of each Begomovirus was analyzed by host species. Among the 16 samples collected, RhGMCV was the most prevalent virus (31.25%), followed by PRMV (25%) and PLDV (12.5%). PYMV and CroGMV were not detected. Corchorus orinocensis was found to be singly infected with RhGMCV, marking, to the best of our knowledge, the first report of this species as a host for Begomovirus. Tithonia diversifolia, Malvastrum coromandelianum, and Solanum nigrum were also identified as new reservoirs in Colombia, each exhibiting single infections with RhGMCV, PRMV, or an uncharacterized Begomovirus. Macroptilium lathyroides presented a mixed infection with PRMV, PLDV, and RhGMCV. These findings expand the known range of natural Begomovirus reservoirs in Valle del Cauca and contribute valuable data for the development of targeted management strategies to mitigate viral disease in crops of agricultural importance.
Non-biological pollination in tomato (Solanum lycopersicum L.) under protected conditions: A systematic review of mechanical and physical methods http://www.scielo.org.co/scielo.php?script=sci_arttext&pid=S2011-21732025000300009&lng=pt&nrm=iso&tlng=pt ABSTRACT In protected horticultural systems, tomato (Solanum lycopersicum L.) reproduction is limited by low biological pollinator activity. To address this issue, non-biological pollination techniques based on physical and mechanical stimuli have been developed. This systematic review analyzed the principles, applications, and agronomic outcomes of these technologies. A systematic review was conducted according to the PRISMA 2020 guidelines. Literature searches were performed between March and April 2024 in seven academic databases (Scopus, ScienceDirect, SpringerLink, MDPI, ProQuest, SciELO, and Google Scholar) with no restrictions on the language. Studies published from 1990 to 2024 that evaluated non-biological pollination in protected tomato systems and reported agronomic variables were included in this review. Predefined inclusion and exclusion criteria were applied. Data on the methods, application parameters, and agronomic outcomes were then extracted. A total of 34 studies were included. Vibration-based techniques (manual, structural, acoustic, or airflow-assisted) have positive effects on fruit set, size, and fruit quality. Devices such as electric brushes, air blowers, and tuning forks are effective and affordable. Emerging technologies, such as flower-recognition drones and aerodynamic guidance systems, have further improved the efficacy of these methods. Growth regulators are useful under environmental stress, although the results vary by cultivar and context. Non-biological pollination methods have improved reproductive efficiency, fruit uniformity, and adaptability, and reduced operational costs. Their effectiveness depends on environmental (temperature, humidity, and airflow) and genetic factors (style length and self-compatibility). The integration of these methods requires site-specific adjustments to optimize agronomic performance.
Physicochemical and functional characterization of melloco mucilage (Ullucus tuberosus Caldas) and its possible agroindustrial use http://www.scielo.org.co/scielo.php?script=sci_arttext&pid=S2011-21732025000300010&lng=pt&nrm=iso&tlng=pt ABSTRACT Melloco, an Andean tuber primarily cultivated by small farmers, has historically been undervalued in the food industry despite its great potential. This study focused on the physicochemical and functional characterization of mucilage extracted from three melloco varieties (INIAP-Caramelo, INIAP-Puca, and INIAP-Quillu) using microwave- and ultrasound-assisted extraction methods. Reducing sugars, total and soluble proteins, amino acids, emulsifying capacity, flocculating capacity, viscosity, and saponin content were evaluated. Significant differences between treatments, both the melloco variety as the extraction method. INIAP-Puca×ultrasound presented the highest values for reducing sugars (3,473.63±0.60 mg L-1) and soluble proteins (8.37±0.00262 mg mL-1); INIAP-Quillu×microwave recorded the highest viscosity (7.97±0.59 cP); INIAP-Puca×ultrasound stood out in saponin content (39.69±4.54 mg/100 g of mucilage); while INIAP-Caramelo×microwave obtained the highest value for total proteins (3,243±953 mg mL-1). Additionally, INIAP-Quillu×ultrasound showed the best results in emulsifying capacity (30.00±3.00%) and turbidity reduction rate (7.66±0.75%). The ultrasound method and the INIAP-Quillu and INIAP-Puca varieties presented best results. These results highlight the great potential of melloco mucilage as a stabilizer and texturizing agent in agro-industrial products, such as sauces, ice creams, dressings, jellies, and beverages, opening new opportunities for the development of functional foods and innovative products.
Current strategies for managing Xylella fastidiosa (2019-2025): A systematic review http://www.scielo.org.co/scielo.php?script=sci_arttext&pid=S2011-21732025000300011&lng=pt&nrm=iso&tlng=pt ABSTRACT Xylella fastidiosa (Wells et al.) represents a major global phytosanitary threat due to its broad host range, rapid dispersal, and frequent asymptomatic infections, which complicate early detection and facilitate pathogen spread. Early and accurate diagnosis is therefore critical as a foundational component of effective management, enabling timely intervention, resource prioritization, and minimization of unnoticed disease dissemination. This review systematically analyzed literature published between 2019 and 2025 in Scopus, Web of Science, and Google Scholar, selecting 50 studies addressing integrated management strategies classified into: (i) diagnostic tools and early detection, (ii) vector control, particularly of Philaenus spumarius, (iii) chemical and nanotechnological strategies, (iv) natural compounds and plant-based approaches, and (v) genetic resistance and varietal improvement, with special emphasis on the Colombian context. Results indicate that while chemical and nanotechnological optimizing strategies have received the greatest research attention, the integration of early diagnostic tools is essential for all other control measures. Vector management and genetic resistance complement these actions, and natural compounds emerge as promising sustainable alternatives. Overall, no single strategy is sufficient to eradicate X. fastidiosa. The most effective approach relies on combining early and accurate diagnosis with complementary control tools that balance scientific efficacy, environmental sustainability, and economic feasibility. This structured framework supports evidence-based decision-making and guides the implementation of integrated management strategies in regions under phytosanitary risk, including Colombia.
