Abstract

Smoking is an important cause of morbid-mortality around the world. Nicotine has stimulant effects and is responsible of cigarette dependence. Main brain structures involved in nicotine dependence are prefrontal cortex, accumbens, hippocampus and amygdala. Nicotine acts both, directly and indirectly on dopamine, acetylcholine, endogenous opioids, noradrenaline and serotonin. Nicotine abstinence is characterized by severe anxiety, craving for tobacco, irritability, restlessness, attention and concentration troubles, increase appetite, and weight, insomnia, headache, and dysphoria or depressive mood. Pharmacologic treatment of nicotine dependence includes either nicotine replacement or medications, such as anxiolytics, alpha-2-adrenergic, nicotine agonist or antagonist, and opiote antagonist.
