Objetive.
To develop and evaluate primers to detect the Protamine 3 (PRM3) gene in alpacas.
Materials and methods.
To do this, the candidate primers were designed based on the conserved sequences obtained from the alignment of the protamine 3 (PRM3) gene with the predicted sequence of the mRNA, both present in the NCBI database. For the in silico evaluation, the primers were compared with the alpaca genome. For in vitro evaluation, 51 testicular tissue samples were used.
Results.
10 pairs of primers were obtained. Of these, pairs 1 and 6 were selected; named PRM3-P31 and PRM3-P32 and with amplicons of 240 bp and 328 bp, respectively. Furthermore, the evaluation of the presence of homodimers and heterodimers was carried out using the Oligoanalyzer program.
Conclusions.
It was concluded that the primer PRM3-P31 allows us to determine the presence of the gene safer than other selected primer. Therefore, it was possible to develop and evaluate primers to detect the presence of the protamine 3 gene in alpacas.
