Summary The objective of this study was to compare two methods (precipitate vs. direct) to quantify low-density lipoprotein cholesterol in laying hens. Materials and methods: Cholesterol in low density lipoproteins was quantified using the precipitate (polyvinyl sulfate) method and the direct method based on detergent and N,Nbis (4-sulfobutyl )-m-toluidine. A total of 40 laying hens (Hy -Line W-36) aged 26 weeks were used. Results: Cholesterol levels (mg/dL) assessed by the precipitate and direct methods were 117.3 ± 38.6 and 49.3 ± 11.1, respectively. The P value for the F test was less than 0.05, showing statistically significant difference between methods, with a 95% confidence. Conclusions: The direct method based on detergents and N,Nbis(4-sulfobutyl)-m-toluidine can be used to determine cholesterol in low density lipoproteins of laying hens since it is not affected by serum triglyceride levels.
