Abstract

From investigation in the 60's and therapeutic response to tricyclic antirepressants, naradrenaline has been involved in the etiology of major depressive episodes. Introduction of selective serotonin reuptake inhibitors, equally efficientbut with lesser incidence of secondary effects than tricyclics, favored serotoninergic etiology in the affective disorders. Recent investigations and synthesis of antidepressive molecules, such as reboxetine, selective noradrenaline reuptake inhibitors (NARI), newly emphasizes the role of noradrenaline in etiology and therapy of depressive episodes. This paper reviews biomolecular aspects, animal investigation models and pharmacodynamics of noradrenergic antidepressive molecules.
