Resumen

Objective:

To use gas chromatography with a flame ionization detector (GC FID) to establish a protocol for quantification of the in vitro production of methane present in the ruminal fluid of Bos taurus.

Methodology:

To establish the protocol for the quantification of the in vitro production of methane present in the ruminal fluid of Bos taurus. The standardization of the chromatographic analysis method for the quantification of CH4 was carried out following the following steps: a) creation of the of the method conditions; b) preparation of the standard concentrations; c) creation of the calibration curve; d) recognition of the retention time; and e) application of the method.

Results:

Regarding the results of the application of the method, the coefficient of variation was less than 15% established by international standards and described as a limit for the adequate validation of an analytical procedure. This indicates that the degree of concordance obtained from the sample replicates is high and the analyzes are repeatable.

Conclusion:

Methane levels can increase or decrease depending on the diet provided to Bos taurus. In this sense, if a correct application of the protocols is carried out, the technique allows quantifying in an effective, reliable and reproducible way the amount of methane gas produced in vitro.
