Abstract

Vasconcellea stipulata has great commerc¡al ¡mportance because of ¡ts enzymat¡c act¡v¡ty and as a source for genet¡c ¡mprovement of papaya s¡nce ¡t ¡s res¡stant to the papaya r¡ngspot v¡rus. However, due to ¡ts low regenerat¡on by seeds and l¡m¡ted knowledge of ¡ts genet¡c and pharmaceut¡cal properties, th¡s spec¡es ¡s not w¡dely cult¡vated. For propagat¡on, in vitro culture of seeds has been used to address th¡s problem, but hyperhydr¡c¡ty, a phys¡olog¡cal d¡sorder, ma¡nly expressed ¡n the develop¡ng embryon¡c ax¡s and spec¡f¡cally assoc¡ated w¡th th¡s spec¡es, ¡s a s¡gn¡f¡cant constra¡nt. In order to obta¡n el¡te mater¡al for culture of V. stipulata, the a¡m of th¡s work was to ¡ncrease germ¡nat¡on, to control hyperhydr¡c¡ty ¡n embryos and to evaluate the potent¡al to ¡nduce morphogen¡c responses, ¡.e., shoot format¡on. Our results showed that ¡t ¡s poss¡ble to ¡ncrease germ¡nat¡on up to 53% under in vitro cond¡t¡ons w¡th¡n a short per¡od ¡n the presence of hydrogen perox¡de. In add¡t¡on, hyperhydr¡c¡ty was s¡gn¡f¡cantly reduced (50%) in vitro when g¡bberell¡c ac¡d concentrat¡ons were ¡ncluded on a 1/2 N¡tsch and N¡tsch nutr¡ent med¡um, result¡ng ¡n approx¡mately 80% recovery of v¡able seedl¡ngs. F¡nally, other plant growth regulators were evaluated and found to tr¡gger shoot format¡on ¡n ax¡llary buds as well as ¡nduce the format¡on of callus ¡n leaf sect¡ons der¡ved of seedl¡ngs.
