Abstract

Here, we describe the extraction, isolation, and purification process to obtain a crude extract of β -D-glucans from the edible mushroom Pleurotus pulmonarius. From a mixture of mycelium and fruiting bodies of the fungus (basidiocarp), we prepared the crude extract from which we extracted β -D-glucan in aqueous and alkaline fractions. The alkaline fraction in the crude extract was purified by dialysis; in the mycelium and fruiting bodies mixture, β -D-glucans were identified using Fourier-transform infrared spectroscopy and 1H and 13C nuclear magnetic resonance, confirming the presence of the characteristic triple helix structure by the congo red spectrophotometric method. The results showed that β -D-glucans in the mushroom represented 32.5% (w/w) and 17.10% (w/w) in the mycelium. Therefore, the methodology used for the extraction was effective in confirming the presence of high percentages of β -D-glucans in P. pulmonarius.
