Abstract

Micro-algae are sources of lipids with the potential to produce biofuels and large-scale dietary supplements. The present study aims evaluates the role of organic solvents of non-polar and polar-protic types (ratio 1: 2) in the extraction and quantification of total lipids and fatty acid methyl esters (FAMEs) of Scenedesmus microalgae. sp. The freeze-dried micro-algae samples were quantified by modified Blight & Dyer (B&D) and pretreated with hydrochloric acid 3 mol.L-1 (with and without HCl). Chloroform was replaced by ethyl acetate, petroleum ether and ethyl ether and methanol by ethanol, Isopropanol and tert-butanol. The FAMEs of the oils of each treatment, with or without addition of HCl, were quantified by gas chromatography. The results showed that the highest extraction of lipids and fatty acids with HCl in the treatments is explained by cell disruption and lipids release. The organic solvents and alcohols presented adequate efficiency in the presence of HCl, but petroleum ether and ethanol presented similar results to the B&D used as reference. In conclusion, the mixtures between the non-polar and protic polar solvents increased the extraction and the yields, especially when the petroleum ether or ethyl ether with ethanol (HCl), providing the unsaturated FAMEs of neutral lipids.
