O presente estudo avalia o papel dos solventes orgânicos de tipos apolares e próticos polares (razão 1:2) na extração e quantificação de lipídios totais e ácidos graxos metil-ésteres (FAMEs sigla em inglês) da micro-alga Scenedesmus. sp. As amostras da microalga liofilizadas foram quantificados os lipídios por Bligh &amp; Dyer (B&amp;D) modificado e pré-tratadas com acido clorídrico 3 mol·L-1 (com e sem HCl). O clorofórmio foi substituído pelo acetato de etila, éter de petróleo e éter etílico e o metanol por etanol, Isopropanol e terc-butanol. Os FAMEs dos óleos de cada tratamento, com ou sem adição de HCl, foram quantificados por cromatografia gasosa. Os resultados evidenciaram que a maior extração de lipídios e ácidos graxos com HCl nos tratamentos é explicada pelo rompimento celular e liberação. Os solventes e álcoois apresentaram eficiência adequada, em presença de HCl, mas éter de petróleo e o etanol apresentaram resultados semelhantes ao de B&amp;D utilizado como referencia. Em conclusão a mistura entre solventes apolares e próticos polares aumentaram os rendimentos, especialmente para quando o éter de petróleo ou etílico misturado com etanol (HCl), disponibilizando os FAMEs de tipo insaturado de lipídios neutros.]]></p></abstract>
<abstract abstract-type="short" xml:lang="es"><p><![CDATA[RESUMEN Las microalgas son fuentes de lípidos con potencial en producir biocombustibles y suplementos alimentarios en gran escala. El presente estudio tiene como objetivo evaluar el papel de los solventes orgánicos apolares y próticos polares (proporción 1:2) en la extracción y cuantificación de lípidos totales y ésteres metílicos de ácidos grasos (FAMEs sigla en inglés) en la microalga Scenedesmus. sp. A las muestras liofilizadas fueron cuantificados los lípidos por Blight &amp; Dyer (B&amp;D) modificado y pre-tratadas con ácido clorhídrico 3 mol.L-1 (con y sin HCl). El cloroformo fue reemplazado por acetato de etilo, éter de petróleo y éter etílico, y el metanol por etanol, isopropanol y terc-butanol. Los FAMEs en los tratamientos, con o sin adición de HCl, fueron determinados por cromatografía de gases. Los resultados evidenciaron que la mayor extracción de lípidos y FAMEs con HCl en los tratamientos se explica por rompimiento de la celular y liberación de los lípidos. Los solventes orgánicos y los alcoholes mostraron una eficiencia adecuada en presencia de HCl, igualmente el éter de petróleo y etanol demostraron resultados similares con B&amp;D utilizado como referencia. En conclusión, la combinación entre los solventes apolares e próticos polares aumentaron la extracción y los rendimientos, especialmente cuando el éter de petróleo o el éter etílico con etanol (HCl), proporcionando los FAMEs insaturados de lípidos neutros.]]></p></abstract>
<abstract abstract-type="short" xml:lang="en"><p><![CDATA[ABSTRACT Micro-algae are sources of lipids with the potential to produce biofuels and large-scale dietary supplements. The present study aims evaluates the role of organic solvents of non-polar and polar-protic types (ratio 1: 2) in the extraction and quantification of total lipids and fatty acid methyl esters (FAMEs) of Scenedesmus microalgae. sp. The freeze-dried micro-algae samples were quantified by modified Blight &amp; Dyer (B&amp;D) and pretreated with hydrochloric acid 3 mol.L-1 (with and without HCl). Chloroform was replaced by ethyl acetate, petroleum ether and ethyl ether and methanol by ethanol, Isopropanol and tert-butanol. The FAMEs of the oils of each treatment, with or without addition of HCl, were quantified by gas chromatography. The results showed that the highest extraction of lipids and fatty acids with HCl in the treatments is explained by cell disruption and lipids release. The organic solvents and alcohols presented adequate efficiency in the presence of HCl, but petroleum ether and ethanol presented similar results to the B&amp;D used as reference. In conclusion, the mixtures between the non-polar and protic polar solvents increased the extraction and the yields, especially when the petroleum ether or ethyl ether with ethanol (HCl), providing the unsaturated FAMEs of neutral lipids.]]></p></abstract>
